HTRF™ Technology: An efficient, wash-free alternative for protein detection

In protein analysis, speed, sensitivity, and reliability of results are essential for advancing research and development. Revvity's HTRF™ (Homogeneous Time-Resolved Fluorescence) technology combines fluorescence resonance energy transfer (FRET) and time-resolved fluorescence measurement to deliver accurate detections without wash steps, reducing costs, time, and errors in complex biomolecular assays.

Traditional methods for protein analysis, such as ELISA assays or protein microarrays, are often time-consuming, involve laborious processes, and have limited sensitivity. Furthermore, the need to divide the sample into multiple aliquots and perform individual tests for each protein significantly increases costs and the risk of error.

What is HTRF™ technology?

HTRF™ (Homogeneous Time-Resolved Fluorescence) is a biomolecule detection technology that offers a homogeneous, sensitive, and efficient solution for analyzing molecular interactions. Its main advantage is that it requires no washing or separation steps, making it an ideal choice for high-throughput environments and laboratories seeking to save time and resources without compromising data quality.

How does HTRF™ work?

HTRF™ combines two key principles:

  1. Fluorescence resonance energy transfer (FRET) : This technique is based on the transfer of energy between a donor fluorophore and an acceptor when they are in close proximity. When the donor is excited, it transfers energy to the acceptor, which emits a specific fluorescent signal if there is an interaction between the molecules of interest.
  2. Time-resolved fluorescence measurement : After excitation, a short delay is introduced before measuring the signal. This allows unwanted background fluorescence (from the medium, free proteins, or interfering compounds) to dissipate, improving accuracy and reducing noise.

Advantages of using HTRF™ in biomolecular assays

  • Quick and easy to use : no centrifugation, washing, filtration or magnetic separation required.
  • Signal stability: The fluorescence signal is stable and not sensitive to light, allowing for repeated readings and greater reliability.
  • High sensitivity and specificity : ideal for detecting protein-protein or ligand-receptor interactions.
  • Time and cost savings : allows multiple analyses in parallel with a single sample.
  • Ideal for screening and complex cell studies : widely used in pharmaceutical research, biotechnology and drug development.
  • Versatility of applications: useful in basic research, target discovery and validation, HTS, primary and secondary screening…

Why adopt HTRF™ in your laboratory?

If you're looking for a protein detection technology that combines speed, reliability, and ease of use, HTRF™ is a safe bet. Its homogeneous format and ability to eliminate interferences ensure more consistent results, even with complex samples.
